In biological analysis, the concentration of the tested compound usually refers to the concentration in the plasma. For most compounds, there is no significant difference in their concentration in plasma compared to that in blood. However, it is possible that plasma concentrations may differ from blood concentrations because compounds may selectively bind proteins in blood cells. Therefore, testing the blood/plasma ratio of a compound can help to understand whether it enters and binds to proteins in blood cells. If the blood/plasma ratio is greater than 1, it indicates that the compound has entered the blood cells, and the PK parameters calculated using serum concentration will not reflect the PK properties of the compound well.
